    K-On! bercerita mengenai empat siswi Sekolah Menengah Atas Sakuragaoka di Jepang. Sebagai kegiatan ekstrakurikuler, mereka bergabung dalam sebuah klub musik pop yang sudah hampir ditutup. Meskipun hanya mereka anggota dari klub itu, namun pada dasarnya mereka bukanlah orang-orang yang berpengalaman dalam bidang musik. Pada awalnya, Yui Hirasawa tidak memiliki pengalaman bermain alat musik atau membacapartitur. Dia hanya pernah bermain kastenyet, tapi akhirnya dia mahir bermain gitar. Sejak itu pula, Yui bersama pemain bass Mio Akiyama, pemain drum Ritsu Tainaka, dan pemain kibor Tsumugi Kotobuki menghabiskan waktu seusai pelajaran sekolah dengan berlatih, ikut serta dalam pertunjukan, atau sekadar jalan-jalan bersama. Klub ini diawasi oleh seorang guru musik bernama Sawako Yamanaka. Sawako nantinya menjadi wali kelas mereka pada akhir tahun mereka di SMA. Ketika mereka duduk di kelas 2 SMA, klub ini mendapat tambahan gitaris baru, bernama Azusa Nakano. Setelah Azusa bergabung, klub ini makin terstruktur dan lebih sering berlatih.

     Pada akhir tahun ketiga, anggota senior klub ini, Yui, Ritsu, Mio, dan Tsumugi lulus SMA, dan mereka setuju untuk masuk ke universitas yang sama. Mereka kemudian bergabung dengan klub musik ringan di universitas bersama tiga teman baru (tiga orang tersebut merupakan satu band). Tiga teman baru mereka bernama Akira Wada (gitar), Ayame Yoshida (drum), dan Sachi Hayashi (bass). Sementara itu, Azusa Nakano yang masih duduk di kelas 3 SMA meneruskan klub musik ringan yang ditinggalkan oleh seniornya (Yui, Mio, Ritsu, Tsumugi). Azusa kemudian menjabat sebagai ketua klub, bersama Ui Hirasawa (adik perempuan Yui) sebagai gitaris dan Jun Suzuki (bass).
